(1) 表观组学实验技术与计算工具开发
以纳米孔测序(nanopore sequencing)为代表的第三代“单分子、长读长”测序技术(简称“三代测序”)以其长读长(典型10-50 kb,可达2 Mbp)、单分子(可用于定量分析)和本源测序(可直接检测修饰碱基)的优势,正在重塑组学研究。我们将开发基于三代测序的表观组学(包括表观基因组和表观转录组)实验技术与计算工具,在单分子和长尺度水平定量地研究基因调控规律。
(2) 生殖细胞和早期胚胎发育研究
生殖细胞发育和早期胚胎发育是表观遗传信息发生全局性重编程的两个关键窗口期,对遗传信息的跨代传递至关重要。我们将利用开发的组学技术与工具,系统探究生殖细胞和早期胚胎发育的调控机制。重点关注的事件包括但不限于:DNA甲基化(DNA methylation)重塑、母源RNA降解(maternal RNA decay, MD)、合子基因组激活(zygotic genome activation, ZGA)、转座元件(transposable element, TE)激活、等位基因特异性表达(allele-specific expression, ASE; 如印记基因 imprinted gene)。
